Sexually Dimorphic Effects of Maternal Alcohol Consumption and Adrenalectomy on Left Ventricular Hypertrophy in Rat Offspring

چکیده
E-00552-2002.R2 In humans, low birth weight and increased placental weight can be associated with cardiovascular disease in adulthood. Low birth weight and increased placental size are known to occur after fetal alcohol exposure or prenatal glucocorticoid administration. Thus, the effects of removing the alcohol-induced increase in maternal corticosterone by maternal adrenalectomy on predictors of cardiovascular disease in adulthood were examined. Alcohol exposure of dams during the last two weeks of gestation resulted in significantly decreased fetal weight and increased placental weight on gestational day 21. Adult female, but not male, offspring of alcohol-consuming mothers exhibited left ventricular hypertrophy. Placental 11β-hydroxysteroid dehydrogenase-2 (11β-HSD-2) mRNA levels, measured by Northern blot, were decreased in females but not males. Adrenalectomy of alcohol consuming dams reversed the increase in placental weight and the decrease in female placental 11β-HSD 2 expression, and eliminated the left ventricular hypertrophy of adult female offspring. These data suggest that alcoholinduced changes in placental 11β-HSD-2 mRNA levels and left ventricular weight are coupled in female offspring only, and depend on maternal adrenal status.

متن کاملLow‐dose maternal alcohol consumption: effects in the hearts of offspring in early life and adulthood
High alcohol consumption during pregnancy leads to deleterious effects on fetal cardiac structure and it also affects cardiomyocyte growth and maturation. This study aimed to determine whether low levels of maternal alcohol consumption are also detrimental to cardiomyocyte and cardiac growth in the early life of offspring and whether cardiac structure and function in adulthood is affected. Preg...
متن کاملStereological Study on the Effects of Maternal Caffeine Consumption on Histomorphometric Changes of Testis and Prostate in Rat Offspring
Background and Objective: It is conceivable that caffeine consumption would induce gonadal changes. The aim of this study is to assess the impact of embryonic caffeine exposure on rat testis and prostate. Materials and Methods: Female rats were divided into (n=7): A control, only received drinking water. B and C groups received caffeine low dose (26 mg/kg) and high dose (45 mg/kg) respectively...
متن کاملMaternal adrenalectomy eliminates a surge of plasma dehydroepiandrosterone in the mother and attenuates the prenatal testosterone surge in the male fetus.
Previous work has established a number of sex-related deficits in immune function, behavior, and endocrine responses to stress in the offspring of dams exposed to ethanol. To examine the potential role of maternal glucocorticoids as a mediator of these sexually dimorphic effects in the fetus, we examined the influence of prenatal alcohol exposure in the presence or absence of maternal glucocort...
متن کاملResveratrol Suppresses Cardiac Renin Angiotensin System in the Late Phase of Left Ventricular Hypertrophy
Background and objectives: Resveratrol(3,5,4′-trihydroxy-trans-stilbene) is a natural polyphenole phytoalexin which exerts potential cardioprotective effects, but the cellular and molecular mechanisms responsible for these effects are still unknown. Cardiac renin angiotensin system (RAS) over-activation plays an important role in pathogenesis of left ventricula...
